Antigen Analysis of Rickettsia typhi Isolated in Korea-SDS-PAGE and Immunoblotting Characters-

Abstract
Antigens of R. typhi(87-91, 87-100) isolated in Korea and reference strain of Rickettsia typhi (Wilmington ATCC VR-144) were characterized by SDS-PAGE and immunoblotting with monoclonal antibodies. The isolates(87-91 and 87-100) were identical
with
reference strain of R. typhi(Wilmington ATCC VR-144) serologically and pathologically. The isolates demonstrated almost same pattern of polypeptide in SDS-PAGE and immunoblotting in comparison with reference strain.
In the SDS-PAGE, the purified antigen of R. typhi isolates demonstrated as many as 10 polypeptide bands corresponded to molecular size 135Kd, 129Kd, 105Kd, 75Kd, 64Kd, 52Kd, 47Kd, 22Kd, 19Kd.
In the immunoblotting, about 10 of polypeptides between 28Kd showed major antigenic activities with hyperimmune sera of BALB/c mice against R. typhi. The strong reactive band were 135Kd, 109Kd, 64Kd, 53Kd, 47Kd, 28Kd.
Antigens and immunoglobulin classes involved in the human antibody response to natural infections with R. typhi were also investigated by western blotting. The result indicated that 135Kd, 100Kd, 97Kd, 75Kd, 50Kd peptides may play a role the most
potent
antigen inducing the antibody in the infections.
